Lab Work

The lab experiment was carried out at Bath Spa University, 2014. 

All of the real example images included in this tutorial were taken in the lab experiment presented in this tutorial.

 

 

Materials

  • Microscope                                                                                

  • Microscopic Camera                                                                   

  • Prepared Blood Smear Slides (10 Slides)                                 

  • Latest ImageJ Sotftware(available at http://imagej.nih.gov/)

Procedure

This lab procedure follows the same structure of one using a miscroscope on histological samples. The only addition is to take images in that process. Several key points in the procedure are mentioned below. 

 

  • Ensure the nosepiece is at 40x magnification when taking the image as this will affect the pixel/scale ratio in the analysis stage.                                                                                              

  • Using the miscroscopic camera, obtain a clear image of dense red blood cells from each prepared blood smear slide (10 in total).

 

Examples of Images Taken 

Figure 1. Four images of red blood cells taken from the microscopic camera. The obtained images have parts with discoloration, which may be caused by several small stains on the microscope's lens or microscopic camera's lens. This problem has to be dealt with in the image processing stage.
